Position Summary
The Senior Research Professional/Lab Manager will assist the PI and laboratory in various aspects of research including experimental laboratory work, data analysis, data preparation and presentation, project coordination, animal husbandry and colony management, laboratory supply inventory/management, maintaining lab SOP and protocols, and training new lab personnel. 50% of the individual's time will be allocated to ongoing research projects in the lab. 25% of the individual's time will be allocated to establishing and maintaining a mouse colony for two individual labs: for the Ananth laboratory and the Kim laboratory. 15% of the individual's time will be allocated to coordination and management of laboratory resources. 10% of the individual's time will be allocated to training/supporting student researchers and technicians.

Experimental:
A minimum of 5 years of training and experimental experience is needed to perform this array of experimental procedures

Performs simple and experimental surgeries on animals (3-5 years of training and experimental experience to perform this array of experimental surgeries)

  • Mastery of animal surgical techniques including: neonatal transverse sinus injection, mouse stereotaxic injections, jugular vein catheterization including ongoing post-operative care
  • Post operative care
  • Handle, dissect and euthanize animals
  • Perform skilled routine surgical procedures in support of on-going animal studies.


Performs animal behavioral experiments (3-5 years of training and specialized experimental experience to master this array of techniques - these skills are specifically required for the Ananth Lab's Research Program)

  • Habituation of animals prior to experimental procedures (contextual habituation, handler habituation, injection habituation)
  • Mouse innate and learned threat assays (cued fear conditioning, odor preference), exploratory assays (open field, displaced object recognition, novel object recognition, conditioned place preference), mouse drug administration (operant conditioning)
  • Analysis of mouse behavior using Noldus Ethovision and/or other open source behavior tracking programs

Prepares animal tissues for fresh and fixed applications (2-3 years of training and experimental experience)

  • Collection of animal tissue for genotyping/colony maintenance/experimental designation ear punch, ear tag, toe clip
  • Harvest and collect animal embryos, animal tissues, and organs for experiments and biopsy for fresh tissue applications.

Performs molecular biology techniques (*3-5 years of training and specialized experimental experience to master this array of techniques - these skills are specifically required for the Ananth Lab's Research Program)

  • Perform a variety of specialized tissue fixation, embedding, sectioning and staining techniques.
  • Perform histological analysis of animal tissues (both cryo-preserved slices AND fresh-prepped slices) stained to demonstrate normal and abnormal structures (Cresyl Violet, H/E, Nissl)
  • Perform fresh tissue collection for subsequent cell/nuclei isolation and RNAseq or mFISH assays.
  • Perform DNA isolation for subsequent genotyping
  • Design oligos and touch-down PCR protocols for ampliation of DNA for genotyping
  • Perform gel-electrophoresis
  • Rapid isolation of anatomically distinct regions of mouse brain for subsequent transcriptomics
  • Sucrose density gradient and nuclei isolation for subsequent single cell RNAseq
  • Perform RNAseq library preparation (10x genomics library preparation)
  • Perform RNAseq library QC for subsequent sequencing (Qubit analysis, Tapestation analysis, miseq QC sequencing to verify capture).
  • Performs RNAscope (mFISH) for multi target labeling of mRNA targets

Performs immunohistochemistry on animal tissues

  • *Use of antibodies and target specific labels to label targets in mouse, nhp and human tissue (fresh floating and affixed to slide) including experimental planning for optimal imaging output
  • *Perform light and confocal microscopy (epifluorescent scope, slide scanner platform, laser scanning confocal) while preserving tissue quality. Up to five co-labeled color imaging

Analyzes experimental data (2-3 years of experience and specialized experience to master this - these skills are specifically required for the Ananth Lab's Research Program)

  • Update and maintain laboratory notebooks and databases; collect, analyze and interpret data.
  • Maintain records of experimental results;
  • Prepare data visualization using a variety of specialized programs: R-studio, graph pad, Loupe browser, Olyvia, IMARIS
  • Quantitative image analysis using IMARIS and Image J including region specific automated cell counting of multi-labeled cells, colocalization analysis
  • Perform statistical analysis of different types of data

Edits and writes research papers and protocols and communicates scientific findings

  • Document information on procedures, modifications and methods for input into final publication.

Duties/Responsibilities
Colony Maintenance:
these tasks will be performed for BOTH the Ananth and Kim laboratories. Five years of specialized animal health training/experience in animal care facilities; ALAT certification required:

  • Studies the basic principles of animal life and the effects of varying environmental factors.
  • Coordinate husbandry, animal care and enrichment activities to maintain a clean, safe and regulatory compliant animal research program.


Conducts research and performs experiments on the selection and breeding of animals.

  • Maintain, breed, genotype and phenotype transgenic animal colony; maintain records of animal colony.
  • Develop and maintain animal breeding records; record lineage, genotype, birth, death and experimental use of each animal.
  • Set up breeding so animals are available at the time required.
  • Develop and maintain a breeding database to record all animals in the breeding colony.
  • Anticipate animal needs for lab research and colony maintenance; adjust the number of breeding pairs accordingly.

Maintains animal colonies.

  • Communicate and collaborate with lab members regarding colony and support duties.
  • Handle, dissect and euthanize animals.
  • Maintain animal colony, including general husbandry, breeding, plug checks, weaning, tagging animals and obtaining tail or ear snips for genotyping, and euthanasia.

Duties/Responsibilities
Lab Management/Training (2-3 years of experience; specific experience working with various safety protocols and training students):

  • Modification of laboratory protocols and experiments based on literature findings.
  • Document all laboratory procedures, experiments and results; maintain and update laboratory notebooks and databases.
  • Develop, validate, perform and interpret a variety of complex experimental techniques and procedures (see above); work with staff to develop procedures addressing new animal study needs, logistics, modifications to standard operating procedures, and/or existing guidelines to resolve technical problems.
  • Manage oversight and progress of critical results in animal-based experiments including evaluating goal-specific phenotypes of animals; collaborate with experimental team members to determine, evaluate and discuss the progress of projects.
  • Keep updated records of lab consumables, order replacement items when needed, communicate Purchasing Agents/Shipping Dock Master as needed to ensure all items are ordered, delivered, and received.
  • Obtain quotes from vendors, communicate with lab members to place orders for large/specialty items, complete and submit various order documents (i.e., market research document, SOW, etc.), and coordinate the shipment of items from the lab to vendors/other institutions.
  • Keep track of past and current order details and regularly update lab on status of orders
  • Flush eyewashes weekly, properly discard of and replace lab waste bins (MPW boxes, sharps containers, broken glass boxes, used pipette containers, general lab recycling), and coordinate the pick-up and drop-off of used/new chemical waste containers.
  • Make sure the lab is up-to-date and up to regulations for various inspections, including OHS, ESH, IACUC protocols
  • Review chemical storage, proper logs, labeling of reagents, etc.; answer questions that inspectors may raise and work with lab to address and correct any potential issues
  • Collect, wash, and autoclave all of lab's used glassware and surgical tools, and restock shelves with clean items weekly.
  • Coordinate events like equipment service, re-certification, preventative maintenance, and installation and communicate with service technicians to ensure lab equipment is serviced and up to operating standards.
  • Support lab personnel with protocol development and troubleshooting and train on proper use of laboratory equipment, experimental methods, proper laboratory techniques, and appropriate use of laboratory resources.
